In general, tube upgrades can have an impact on the sound quality and performance of tube-based amplifiers. Upgrading the power tubes can potentially result in improved clarity, warmth, dynamics, and overall tonal characteristics. However, the effect of tube upgrades can vary depending on the specific device, the quality of the stock tubes, and personal preferences.
Sorry, I know criticizing misspelling is poor manners on the Internet, but I can't resist highlighting DUCK Tubes.
Also - the question must be asked, is the Little Dot capable of responding to small quality differences in tubes? Some amps are, some aren't. Power tubes will also have very little effect on sound quality versus the pre-amp tubes. You've already upgraded the pre-amp tubes (with great Duck tubes!). You might be better off to look elsewhere for improving sound than the power tubes.
